Acid mucopolysaccharides in metal-induced tumor and connective tissue capsules surrounding implant in rats

Abstract
A study was made of changes of acid mucoplysacharides in a malignant tumor and connective tissue capsules following a subcutaneous imbedding of four metal plates in Sprague-Dawley rats, in an attempt to clarify the induction of tumor in reference to the histological findings and the components of acid mucopolysaccharides. Acid mucopolysaccharides were isolated through a proteolytic digestion from the malignant tumor developed and the connective tissue capsules of four groups 90, 180, 270 and 360 days after imbedding, and fractionated with cetylpyridinium chloride. The components of acid mucopolysaccharides were studied with a two-dimensional electrophoresis on a cellulose acetate strip, hexosamine analysis, chondroitin sulfate isomers analysis, and enzymatic methods with the application of chodroitinase ABC and chondroitinase AC.
The results obtained were as follows:
1) Fibroblastic activity in the superficial layers of connective tissue capsules was found on the 180th day after imbedding, and the activity increased more markedly in the deeper layers on the 360th day after imbedding.
2) The concentration of acid mucopolysaccharides in connective tissue capsules was highest 180 days, intermediate 270 and 360 days. and lowest 90 days after imbedding.
3) The acid mucopolysaccharides in the developed malignant tumor and for kinds of connective tissue capsules proved to be hyaluronic acid, heparan sulfate, chondroitin sulfate A and chondroitin sulfate B. Only the 90-day-old connective tissue capsule additionally contained very small amounts of heparin.
4) Sulfated mucopolysaccharides in connective tissue capsules showed a gradual increase possibly due to a decrease in nonsulfated mucopolysaccharides. The 180-day-old connective tissue capsule showed an increase in the amounts of both chondroitin sulfate A and chondroitin sulfate B, while the 360day-old revealed more increase in chondroitin sulfate A. The main component in the developed malignant tumor proved to chondroitin sulfate A (43. 0%)
5) An Increase in the amounts of chondroitin sulfate A seems to facilitate malignant change of fibroblasts which appeared in the deeper layers of the 360-day-old connective tissue capusule.
